Indications for Use

The Emit® 2000 Vancomycin Calibrators are intended for use with the Emit® 2000 Vancomycin Assay as a reference for use in determining vancomycin levels in human serum or plasma.

Device Story

Emit® 2000 Vancomycin Calibrators are liquid, six-level calibrators used in clinical laboratories to establish reference points for the Emit® 2000 Vancomycin Assay. They consist of vancomycin, buffer, and preservatives in a synthetic matrix. The calibrators are used by laboratory technicians to calibrate the assay, ensuring accurate quantification of vancomycin levels in patient serum or plasma samples. This calibration process is essential for healthcare providers to monitor therapeutic drug levels and make informed clinical decisions regarding vancomycin dosing.

Clinical Evidence

No clinical data. Bench testing only. Stability was evaluated by comparing calibrators stored at 9°C versus -20°C, supporting a shelf life of at least 13 months. Calibrator lots are validated by testing 20 replicates at each level and comparing values to a Master Lot to ensure consistency.

Technological Characteristics

Six-level liquid calibrator. Composition: vancomycin, buffer, and preservatives in a synthetic matrix. Preparation: standard gravimetric procedures. Calibration: lot-specific adjustment based on 20-replicate testing against a Master Lot. Stability: 13-month shelf life supported by real-time data.

Indications for Use

Indicated for use as a reference standard for determining vancomycin levels in human serum or plasma samples.

Regulatory Classification

Identification

A clinical toxicology calibrator is a device intended for medical purposes for use in a test system to establish points of reference that are used in the determination of values in the measurement of substances in human specimens. A clinical toxicology calibrator can be a mixture of drugs or a specific material for a particular drug (e.g., ethanol, lidocaine, etc.). (See also § 862.2 in this part.)

Special Controls

*Classification.* Class II (special controls). The device is exempt from the premarket notification procedures in subpart E of part 807 of this chapter subject to the limitations in § 862.9.

Emit® Vancomycin Calibrator (K881040) #### Device Description: 4. Emit® 2000 Vancomycin Calibrators are liquid, six-level calibrators prepared from vancomycin, buffer and preservatives. The calibrators are intended for use with the Emit® 2000 Vancomycin Assay as a reference for use in determining vancomycin levels in human serum or plasma. #### Device Intended Use: 5. The Emit® 2000 Vancomycin Calibrators are intended for use with the Emit® 2000 Vancomycin Assay as a reference for use in determining vancomycin levels in human serum or plasma. Medical device to which equivalence is claimed and comparison information: 6. The Emit® 2000 Vancomycin Calibrators are substantially equivalent in intended use to the Emit® Vancomycin Calibrators (K881040) currently marketed. The modified Emit® 2000 Vancomycin Calibrators contain the same vancomycin concentrations as the currently marketed device. Both devices are intended for use in the calibration of human serum or plasma in conjunction with a Vancomycin Reagent. #### 7. Device Performance Characteristics: ## Stability: Shelf life stability was evaluated by testing the calibrators at a minimum in duplicate, stored at 9°C, with comparison to calibrator sets stored at -20°C. The assignment of shelf life will be based on real time data from at least 3 lots of calibrators. To date, stability studies support no significant change in recovery for at least 13 months. ### Preparation: Emit® 2000 Vancomycin Calibrators (modified) are prepared by standard gravimetric procedures using a synthetic matrix with buffers and preservative which has been supplemented with commercially available vancomycin. Each new calibrator lot is tested in bulk and final container stages with 20 replicates at each calibrator level. These values are compared to a Master Lot. Calibrator levels are adjusted as needed to match Emit® 2000 responses of the Master Lot calibrators. {1}------------------------------------------------ ## DEPARTMENT OF HEALTH & HUMAN SERVICES Image /page/1/Picture/1 description: The image shows the seal of the Department of Health & Human Services - USA.
