Intended Use

Arctic™ Sperm Cryopreservation Medium is intended for use in assisted reproductive procedures involving the cryopreservation and storage of human sperm.

Device Story

Arctic™ Sperm Cryopreservation Medium is a defined, single-use chemical medium used in clinical assisted reproductive technology (ART) laboratories. It facilitates the cryopreservation and storage of human sperm samples. The medium contains salts, buffering agents (including MOPS), cryoprotectants, vitamins, amino acids, minerals, carbohydrates, surfactants, energy sources, and human serum albumin. In practice, clinicians mix donor semen with the medium before cryopreservation in liquid nitrogen. The medium protects sperm cells during the freezing and thawing process, maintaining motility for subsequent use in reproductive procedures. The device is supplied in 5 mL sterilized vials.

Clinical Evidence

Bench testing only. Performance validated via pH, osmolality, and sterility testing (USP <71>). Endotoxin testing (USP <85>) confirmed levels ≤1.0 EU/ml. Sperm cryosurvival assay compared subject medium to predicate using donor semen; success defined as ≥80% of control motility post-thaw, post-gradient separation, and two hours post-wash. Accelerated shelf-life testing confirmed stability over 18 months.

Technological Characteristics

Defined medium containing salts, MOPS buffer, cryoprotectants, vitamins, amino acids, minerals, carbohydrates, surfactant, energy source, and human serum albumin. Osmolality: 2300-2600 mOsm/kg; pH: 7.25-7.54. Aseptically filled into sterilized vials (SAL 10^-3). Validated per ISO 13408-1:2008 and ISO 13408-2:2003.

Reproductive media and supplement are products that are used for assisted reproduction procedures. Media include liquid and powder versions of various substances that come in direct physical contact with human gametes or embryos (including water, acid solutions used to treat gametes or embryos, rinsing solutions, sperm separation media, supplements, or oil used to cover the media) for the purposes of preparation, maintenance, transfer or storage. Supplements are specific reagents added to media to enhance specific properties of the media (e.g., proteins, sera, antibiotics, etc.).

Special Controls

*Classification.* Class II (special controls) (mouse embryo assay information, endotoxin testing, sterilization validation, design specifications, labeling requirements, biocompatibility testing, and clinical testing). The device, when it is phosphate-buffered saline used for washing, and short-term handling and manipulation of gametes and embryos; culture oil used as an overlay for culture media containing gametes and embryos; and water for assisted reproduction applications, is exempt from the premarket notification procedures in subpart E of part 807 of this chapter subject to the limitations in § 884.9.

The subject and predicate devices have the same or comparable appearance, pH, and osmolality. The only differences in formulation are the replacement of L-glutamine with L-alanyl glutamine, addition of MOPS buffer, and minor modifications in concentrations of certain ingredients in the subject device. The differences noted are minor and do not raise different questions of safety andeffectiveness. In addition to pH and osmolality, the subject and predicate devices have the same acceptance criteria for the sperm cyrosurvival assay and sterility testing, and the subject device has a more stringent acceptance criterion for endotoxin testing (≤1.0 EU/ml vs. ≤3.0 EU/ml). #### VIII. Summary of Non-Clinical Performance Testing: The following studies have been performed to support substantial equivalence to the predicate device: - pH (7.25-7.54) ● - Osmolality (2300-2600 mOsm/kg) ● - Aseptic Processing Validation testing per ISO 13408-1:2008 and ISO 13408-● 2:2003 - Sterility testing (no microbiological growth) per USP <71> ● {6}------------------------------------------------ - Endotoxin testing (≤1.0 EU/ml) per USP <85> o - Sperm Cryosurvival Assay Donor semen was mixed with the test medium and control (predicate device) and cryopreserved in liquid nitrogen. After thawing, sperm motility was determined at three time points: post thaw, post gradient separation, and two (2) hours post wash.
