Indications for Use

Pasco MIC and MIC/ID panels are used for quantitatively measuring the susceptibility of rapidly growing aerobic and facultative anaerobic bacterial pathogens to a battery of antimicrobial agents and determining the biochemical identification of these organisms. The Gram-Negative MIC/ID, the Gram-Negative MIC/ID II and Gram-Positive MIC/ID Panel contain appropriate batteries of antimicrobial agents and biochemical substrates currently used for determining antimicrobial susceptibility and biochemical identification of various organisms.

Device Story

Pasco MIC panels are multi-well plastic microtitre plates containing frozen serial dilutions of antimicrobial agents. Device used in clinical laboratories by technicians. Inoculum prepared from bacterial colonies using saline and adjusted to 0.5 McFarland standard via spectrophotometer or Director™ Inoculation System. Panels inoculated, incubated 16-24 hours at 35°C in non-CO2 incubator. Technicians perform manual readings using indirect lighted background viewer to identify lowest concentration with no visible growth (MIC). Output provides quantitative susceptibility data to assist clinicians in selecting appropriate antibiotic therapy for bacterial infections.

Clinical Evidence

Bench testing only. Performance evaluated using 115 Staphylococcus aureus isolates (77 clinical, 38 challenge). Compared to NCCLS M-7 reference broth dilution method. Results showed 100% Essential Agreement (EA) and 100% Category Agreement (CA). No major or very major discrepancies observed. Reproducibility testing across three sites demonstrated 100% intra- and inter-site reproducibility.

Various concentrations of antimicrobics are dispensed into the Pasco microdilution panels and the panels are then frozen. Inoculum is prepared in saline and equated to a 0.5 McFarland standard. Panels are thawed prior to use, and inoculated with the test organisms. Plates are incubated for 16-24 hours at $35^{\circ}$ in a non- $\mathrm{CO}^{2}$ incubator and are then observed for visible growth or color changes (ID portion). The lowest concentration of each antimicrobial agent with no apparent visible growth of the test organism is recorded as the minimum inhibitory concentration (MIC). Only manual readings are performed using an indirect lighted background viewer. Inoculation procedures include the Direct Turbidity Standard method, with recommendations to use a spectrophotometer to adjust the final inoculum concentration equal to the 0.5 McFarland standard and the Director™ Inoculation System.
