Enzygnost F1+2 (monoclonal) is an enzyme immunoassay (EIA) in microtiter format for plasma samples. Principle: sandwich immunoassay using monoclonal mouse antibodies. Process: F1+2 antigen in sample binds to antibodies on microtiter plate; peroxidase-conjugated anti-human prothrombin antibodies bind to F1+2 determinant; enzymatic reaction with hydrogen peroxide and chromogen; reaction terminated with sulfuric acid. Output: color intensity measured photometrically, proportional to F1+2 concentration, quantified via calibration curve. Used in clinical laboratory settings by trained personnel. Results assist clinicians in evaluating coagulation system activity, assessing thrombosis risk, and monitoring anticoagulant therapy.

Clinical Evidence

Bench testing only. Precision evaluated over 5 days (n=40) with total CVs ranging from 6.8% to 11.8%. Analytical specificity confirmed no interference from bilirubin (up to 60 mg/dL), hemoglobin (up to 600 mg/dL), triglycerides (up to 3000 ng/dL), or rheumatoid factor (67-197 IU/mL). Method comparison with predicate (n=190) yielded y = 0.265x - 29.378, r = 0.96. Reference range established in 152 healthy subjects (5th-95th percentile: 62-197 pmol/L).

During the first incubation, the F 1+2 antigen in the sample binds to F 1+2 antibodies attached to the surface of the microtitration plate. F1 + 2 concentration is determined from four replicates, to determine if the specification for F1 + 2 concentrations met. If so, the pool is filtered, vialled and lyophilized.
