Intended Use

The UniMark® Home Pregnancy Test Device is intended for the qualitative measurement of human Chorionic Gonadotropin (hCG) in urine as a home use screening test for pregnancy.

Device Story

UniMark® Home Pregnancy Test Device is a rapid, visual, one-step, solid-phase chromatographic immunoassay for qualitative hCG detection in urine. Device consists of a test strip within a plastic cassette. Principle of operation: urine specimen migrates via capillary action through a conjugate pad containing mouse anti-beta hCG antibodies conjugated to colloidal gold particles; hCG in specimen binds to conjugate; complex moves to test zone where immobilized goat anti-hCG antibodies capture the complex, forming a visible purple band. Positive result (purple band) indicates presence of hCG; negative result (absence of band) indicates no detectable hCG. Control window confirms proper reagent performance. Used by consumers at home for early pregnancy screening. Results are visually interpreted by the user to inform pregnancy status.

Clinical Evidence

Bench testing: Sensitivity 25 mIU/ml (WHO 3rd IS 75/537). Accuracy: 364 urine specimens (189 positive, 175 negative) showed 100% agreement with a quantitative visual hCG test. Specificity: No cross-reactivity with hLH (200 mIU/ml), hFSH (1000 mIU/ml), or hTSH (1000 uIU/ml). Interference testing: No interference from common substances (acetaminophen, caffeine, glucose, etc.) at specified concentrations. Consumer study: 100 home users tested coded urine samples (0, 20, 30, 1000 mIU/ml hCG). 100% correlation between home user results and professional laboratory testing.

Performance Characteristics (if/when applicable): 1. Analytical performance: All analytical and method comparison studies were performed previously on the predicate device. Therefore, only the over-the counter lay user study was included in this 510(k). a. Precision/Reproducibility: Qualitative assay: Reproducibility of the device in the hands of lay users was evaluated in a consumer study of 100 users selected on a random basis as they presented themselves at study sites. Participants ranged in age from 16 to over 40 and ranged in education level from high school to graduate levels. Participants were given a set of coded samples containing spiked specimens at concentrations of 0, 20, 30 and 1000 mIU/ml. The only instructions provided were draft copies of the home use package insert. All samples at 30 mIU and 1000 mIU/ml tested positive. Results were compared to those obtained by laboratory professionals. In this study there was 100% agreement between results obtained by lay users and those obtained by professionals. b.
