Immunoassay for the in vitro quantitative determination of Prostate-Specific Antigen in human serum and plasma. The Elecsys PSA assay is further indicated for serial measurement of PSA to aid in the management of cancer patients. The electrochemiluminescence immunoassay "ECLIA" is intended for use on the Boehringer Mannheim Elecsys 2010 immunoassay analyzer.
Device Story
Elecsys PSA Assay is an in vitro diagnostic immunoassay for quantitative PSA measurement in human serum and plasma. Used on the Boehringer Mannheim Elecsys 2010 analyzer, the device employs a sandwich electrochemiluminescence immunoassay (ECLIA) principle. Input consists of 20 µL patient sample incubated with biotinylated monoclonal PSA-specific antibody and ruthenium-labeled monoclonal PSA-specific antibody. Streptavidin-coated microparticles bind the complex to a solid phase. The mixture is magnetically captured on an electrode; unbound substances are washed away. Voltage application induces chemiluminescent emission, measured by a photomultiplier. Results are calculated via a 2-point calibration curve and master curve provided by reagent bar code. The assay aids clinicians in managing cancer patients through serial PSA monitoring. The system is intended for professional use in clinical laboratory settings.
Clinical Evidence
Bench testing only. Performance evaluated via precision studies (N=60 per level), analytical sensitivity (0.002 ng/mL), functional sensitivity (0.03 ng/mL), and linearity (0.002-100 ng/mL). Method comparison against the 1st Gen. predicate (N=108) yielded a correlation of r=0.998 (Passing Bablok: y=1.0335x + 0.219). Hook effect testing showed no interference up to 17,000 ng/mL.
Technological Characteristics
Sandwich electrochemiluminescence immunoassay (ECLIA). Uses biotinylated monoclonal antibodies and ruthenium-labeled monoclonal antibodies with streptavidin-coated microparticles. Energy source: electrical voltage applied to electrode to induce chemiluminescence. Detection: photomultiplier. Connectivity: reagent bar code for master curve input. System: Elecsys 2010 immunoassay analyzer.
Indications for Use
Indicated for in vitro quantitative determination of PSA in human serum and plasma to aid in the management of cancer patients.
Regulatory Classification
Identification
A tumor-associated antigen immunological test system is a device that consists of reagents used to qualitatively or quantitatively measure, by immunochemical techniques, tumor-associated antigens in serum, plasma, urine, or other body fluids. This device is intended as an aid in monitoring patients for disease progress or response to therapy or for the detection of recurrent or residual disease.
Special Controls
*Classification.* Class II (special controls). Tumor markers must comply with the following special controls: (1) A guidance document entitled “Guidance Document for the Submission of Tumor Associated Antigen Premarket Notifications (510(k)s) to FDA,” and (2) voluntary assay performance standards issued by the National Committee on Clinical Laboratory Standards.
