Intended Use

The ARCHITECT B12 assay is a chemiluminescent microparticle Intrinsic Factor assay for the quantitative determination of vitamin B12 in human serum on the ARCHITECT i System. Device Story

The ARCHITECT B12 assay is a two-step chemiluminescent microparticle immunoassay (CMIA) performed on the ARCHITECT i 2000SR instrument. The process involves automated sample pretreatment using sodium hydroxide, potassium cyanide, alpha monothioglycerol, and cobinamide dicyanide to release B12 from binding proteins. Pre-treated samples are combined with intrinsic factor-coated paramagnetic microparticles; B12 binds to the microparticles. After washing, acridinium-labeled B12 conjugate is added. Pre-trigger and trigger solutions initiate a chemiluminescent reaction measured as relative light units (RLUs). The system provides an inverse relationship between B12 concentration and RLUs. The device is used in clinical laboratories by trained personnel. Results assist clinicians in diagnosing and managing anemias related to gastrointestinal malabsorption. Hemolyzed specimens are contraindicated due to negative interference.

Clinical Evidence

No clinical studies were performed. Analytical performance was established via bench testing. Precision was evaluated per CLSI EP5-A2 (N=357, %CV 3.4-5.8%). Linearity was confirmed per CLSI EP6-A (146–2000 pg/mL). Detection limits were established per CLSI EP17-A (LoB 66 pg/mL, LoD 88 pg/mL, LoQ 146 pg/mL). Method comparison with the predicate (N=151) showed a correlation coefficient of 0.991. Interference testing per CLSI EP7-A2 identified negative interference from hemolysis; other substances (bilirubin, protein, triglycerides) showed no significant interference.
