Device Story
NoFact VIII Deficient Plasma is a lyophilized, immunodepleted human plasma reagent used in clinical laboratories to measure Factor VIII activity. The device is mixed with a diluted patient citrated plasma sample and tested using an activated partial thromboplastin time (APTT) assay. The clotting time of the mixture is inversely dependent on the concentration of Factor VIII in the patient sample. The output is a quantitative measurement of Factor VIII activity, which assists clinicians in diagnosing or monitoring Factor VIII deficiency. The device is intended for in vitro diagnostic use by laboratory professionals.
Clinical Evidence
Bench testing only. Performance was evaluated using 233 samples (normal donors and patients with hemophilia, sepsis, VWD, and surgical history) across three sites. Method comparison against the predicate yielded a correlation coefficient (r) of 0.984. Precision studies (n=80 per lot) showed within-device %CV < 10% for normal and < 15% for abnormal controls. Linearity was confirmed from 1% to 160% FVIII activity (r > 0.99). Interference studies confirmed no significant impact from hemoglobin (500 mg/dL), bilirubin (20 mg/dL), or triglycerides (2000 mg/dL).
Technological Characteristics
Lyophilized citrated human plasma immunodepleted of Factor VIII. Measurement principle: APTT-based clotting time assay. Reconstituted stability: 8 hours at 2-8°C, 4 hours at room temperature. Designed for use on automated coagulation analyzers.

- NoFact VIII Deficient Plasma was compared to the predicate STA FVIII Deficient plasma using the ﻘ Stago PTT-A FVIII assay on the STA Compact. Plasma samples were assayed in parallel with the Stago PTT-A assay using the STA FVIII deficient plasma ("Stago results"), and also with the same assay but where the NoFact VIII deficient plasma was substituted for the STA FVIII deficient plasma ("NoFact results"). We have reviewed your Section 510(k) premarket notification of intent to market the device referenced above and have determined the device is substantially equivalent (for the indications for use stated in the enclosure) to legally marketed predicate devices marketed in interstate commerce prior to May 28, 1976, the enactment date of the Medical Device Amendments. or to devices that have been reclassified in accordance with the provisions of the Federal Food, Drug, and Cosmetic Act (Act) that do not require approval of a premarket approval application (PMA).
