Intended Use

QC-Beads™ are intended for use as controls in counting sperm manually using a microscope or automatically using computer assisted semen analysis instruments.

Device Story

QC-Beads™ are quality control materials for sperm concentration analysis; consist of 3.7-μm polystyrene latex beads suspended in glycerol/detergent/water solution with 0.1% sodium azide preservative. Supplied in two concentration levels (Lo: 16-24 million/ml; Hi: 34-46 million/ml). Used by laboratory technicians with manual microscopy (hemacytometers/Makler chambers) or computer-assisted semen analysis (CASA) instruments to verify counting accuracy. Provides a stable, non-biological alternative to human semen samples for daily instrument/method validation.

Clinical Evidence

No clinical data. Bench testing only. Precision/reproducibility evaluated by counting duplicate aliquots of Hi and Lo beads 10 times using a Neubauer hemacytometer and bright-field microscope; results showed CV of 9-11%. Stability testing confirmed at least one-year shelf life at room temperature.

Technological Characteristics

Polystyrene latex beads (3.7-μm diameter) in glycerol/detergent/water solution; 0.1% sodium azide preservative. HDPE bottle packaging. Manual or automated optical counting principle. Room temperature storage.

Indications for Use

Indicated for use as quality control material for sperm counting procedures, including manual microscopic counting and automated computer-assisted semen analysis (CASA) systems.

Regulatory Classification

Identification

A hematology quality control mixture is a device used to ascertain the accuracy and precision of manual, semiautomated, and automated determinations of cell parameters such as white cell count (WBC), red cell count (RBC), platelet count (PLT), hemoglobin, hematocrit (HCT), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC).

Special Controls

*Classification.* Class II (special controls). Except when intended for use in blood components, the device is exempt from the premarket notification procedures in subpart E of part 807 of this chapter subject to the limitations in § 864.9.
